Objective To investage the directional differentiation potential of the embryonic stem cells into pituitary-like cells.Methods 129 mouse embryonic stem cells of E14 were cocultured with human amnion for 4 days to induce the embryonic stem cells of E14 into the epidermal stem cells,then the embryonic stem cells and human amnion were transplanted into 129 mice subcutaneouly by aseptic operation.The differentiation was observed by morphological method after 4 weeek.Results After coculture with amnion for 4d,the epidermal-like stem cells clones were formed on the epithelial surface of amnion with high levels of β1 integrin,CK15 and CK19 expressions.The increasing of the graft was obviously at 4 weeks after ransplantation.There were pituitary-like cells formed in the surface of human amnion.Conclusion The preliminary study suggests that the embryonic stem cells have the differentiation potential to form pituitary-like cells at suitable microenviorment.
