Objective To construct expressing plasmid antisense IGF-1 for gene therapy of rat glioma.Methods IGF-1 cDNA 300 bp were amplified using RT-PCR,sequence.Transfect C6 cells with antisense IGF-1 and the cells proliferative rate and the effect on treatment of C6 glioma were observed in vivo.Results The sequence was correct.The growth of C6 cells transfected was inhibited significantly and tumorigenicity was lost.After treatment,complete disappearance of tumor occured in all tumorbearing rats,and no recurrence during six months.Conclusion Antisense IGF-1 expression plasmid inhibits the growth of C6 cells in vitro,and the treatment is effective in vivo.
