Objective
To study the culture characteristics and temperature sensitivity of varicella-zoster virus (VZV) Oka strain.
Methods
2BS cells infected with VZV Oka strain were cultured. When the cytopathic effect (CPE) of 2BS cells reached 10%, the virus suspensions were harvested at 0, 3, 6, 9, 12, 18, 24, 36 h, and the virus titers were detected. The virus growth curve was observed. The virus protein concentrations were detected daily for 7 d since the first day of VZV culture, and the change of virus protein concentration was observed. The virus suspensions harvested at 3, 12, 24, 36 h were placed at 4 ℃ and 25 ℃ respectively for 0, 3, 6, 9, 12, 24 h, and the virus titers were detected.
Results
The virus growth entered the log phase and plateau phase respectively at 0-12 h and 12-24 h after the CPE of 2BS cells reached 10%, and then entered the decline phase. During VZV culture process, virus protein expression entered log phase at 2-6 d after 2 d initial phase, and then entered the plateau phase. The titer decline magnitude and speed of VZV in different growth periods placed at 4 ℃ were both lower than at 25 ℃.
Conclusion
VZV Oka strain has general virus culture characteristics, and is greatly affected by temperature.
