This review investigates into the topic of purine and pyrimidine derivatives and their potential as anticancer agents. It thoroughly investigates their efficacy against various types of cancer cells and sheds light on the underlying mechanisms at play. The development of these derivatives has sparked immense interest, as they hold great promise for clinical use. They display a remarkable ability to hinder DNA synthesis and repair, as well as regulate the cell cycle, making them powerful weapons against cancer. By the review designed purine derivatives at C2 with C6 and C9 are active against Brest cancer cell lines, C2, C4 and C7 against colon and blood cancer, C4 against kidney cancer cells, C6 against liver, lung, luekamia and colon cancer cell line, C7, Brest, liver and metastatic colon cancer and C8 substituted are active against majority of cancer cell lines. With respect to pyrimidine derivatives tetra substituents at 1, 2, 4 and 6 are active against Metastatic cancer in Brest, pancreas and liver cells, 2, 4 and 6 tri substituted against Metastatic cancer in Brest, pancreas and liver, against Arora kinase, leukemia and colon cancer, Further they are active against non small cell lung cancer and CNS cell lines respectively.
