Abstract In diabetes, AGEs formation and deposition are much enhanced,which has been closely linked to the diabetic chronic complication.However,RAGE mediated the main action for AGEs.RAGE, a new member of immunoglobulin superfamily,is ubiquitously expressed by Mps,Ecs,Mcs,fibroblasts,smooth muscle cells and neurons.RAGE interacts with LF-L noncovalently,and the resulting complex formates the binding sites of AGEs. When binding to it,AGEs can induce oxidant stress to active the signal transduction pathway which was consisted of reactive oxygen intermediates and NF-KB, resulting in changes in gene expression.Thus it can induce the dysfunction of target cells, hence the development of diabetic chronic complications. Therefore, the discovery of RAGE and development of diabetic reagents to block AGEs-RAGE interaction should provide insights into the prevention and therapy of diabetic chronic complications.
