Preparation of bovine serum albumin loaded poly (L-lactic-co-glycolic acid) microspheres and effects of different additives on encapsulation efficiency and release in vitro
Objective To explore effects of different additives and preparation of microsphere on bovine serum albumin loaded poly (L-lactic-co-glycolic acid) (BSA-PLGA) microspheres' encapsulation efficiency and release in vitro. Methods The BSA-PLGA microspheres were prepared by double emulsion-solvent evaporation method. By determining their encapsulation efficiency and releasing in vitro,the used amount of BSA,the concentration of PEG and sodium chloride were studied. Results The preparation technology of microspheres were optimized by orthogonal test. Finally,the BSA-PLGA microspheres ,with the encapsulation efficiency of (91.07 ± 4.22)% and BSA burst release rate of (10.54 ± 3.84)%,could release nearly 28 days in vitro. Conclusion BSA-PLGA microspheres with higher encapsulation efficiency and lower burst release rate could be produced by optimized preparation process and additives.
