Selection on Isolated Species for Bursaphelenchus xylophilus of Inoculationin Resistant-breeding

In order to select the wood nematode resistant alternative pines of Pinus massoniana and to ensure the resources of the pine wood nematode used for inoculation, studies of the reproduction, infectious virulence and propagation have been done since the development of the project in October 2001. Nineteen wilted pine specimens died of nematode in different areas have been collected from which 19 nematode isolated species were separated by means of Baman and finally sixteen isolated species were cultivated successfully under the temperature of 25℃. Then the two years old seedlings of pinus thunbergii were inoculated to examine the infective virulence of the pine wood nematode. The infective rates 55 days later in the field were between 26.1% and 97.6% compared with the rates between 84.4% and 100% in the vinyl house with the weighted averages rate gap higher than 20%, which proved the wood nematode infective virulence strengthened with the temperature increase and the soil dryness. The 16 isolated species with the same number of nematode varied in the speed of the propagation areas in the PDA culture medium. What's more, regarding to mass propagation, the same number of nematode in the same amount of wheat culture medium varied in the quantity of propagation, between 0.98 and 1.76 million per culture dish. According to the result of the experiment, the most virulent and reproductive“Guang Zhu 3B” was selected as the resource of the pine wood nematode for inoculation.
