Experimental Study of Qingwen Baidu Decoction on the Expression of NF-κBp65 of the Acute Iung Injury in Rats

Objective: By observing Qingwen Baidu decoction,the traditional Chinese medicine,on LPS-induced acute lung injury(ALI) lung tissue pathology and the expression of nuclear factor-κBp65(NF-κBp65),and to explore Qingwen Baidu decoction in the treatment of ALI and intervention mechanism.Methods:144 healthy male SD rats were randomly divided into 6 groups(n=24):normal control group,model group,the prednisone group and the Qingwen Baidu decoction high,middle and low dose group.The LPS solution(2mg/kg) was instilled into the tracheal to prepare rat ALI model,once a day for two days.Each treatment group completed the second infusion 1 hour after LPS solution,the drug with saline diluted to the appropriate 4mL to intragastric administration twice a day.After each group were fed 24h,48h,6 rats were killed,all remaining rats were sacrificed at 72h,and to collect the required samples.We observed the pathological changes of lung tissue under light microscope,and detected the NF-κBp65 expression in lung tissue by Western blot.Results: The pathology of rats lung tissue with LPS induced ALI showed the alveolar structure damaged or consolidation,the lung interstitial edema obviously,the alveolar cavity and the lung interstitial were infiltrated by the focal inflammatory cells and red blood cells.Phase model with the same group,the pathological inflammation of rats lung tissue in Qingwen Baidu decoction treatment groups in the integral dose group were significant lower(P0.01),The low-dose group only 24h,48h were significantly or very significantly reduced lower(P 0.05 or P 0.01);The expression of NF-κB p65 in high and middle dose of Qingwen Baidu decoction treatment groups decreased in 48h,and all does treatment groups of Qingwen Baidu decoction decreased in 72h.Compared with the model group phase has significant or very significant difference(P0.05 or P0.01).Conclusion:The Qingwen Baidu decoction can reduce the injured degree of ALI rats tissue,and can reduce the inflammatory cells gather,infiltrate and exudate in the lung tissue,so to play a clearer role in lung protection.The Qingwen Baidu decoction can effectively reduced the express of NF-κB p65 in LPS induced ALI rats tissue.By inhibit the activation of NF-κB and the generation of inflammatory cytokines can reduce the inflammatory response in rats lung tissue.Using the Qingwen Baidu decoction early can effectively reduced lung tissue injured degree and can reduced the inflammatory response in ALI rats lung tissue induced by LPS.
