Physiologically Based Pharmacokinetic Model and Its Applications

The physiologically based pharmacokinetic(PBPK) modeling has become an important direction in drug research and toxicology.Comparing with the conventional compartment pharmacokinetic models,the significant difference is that the PBPK models are 'mechanistically based' mathematical model that can simulate the distribution and metabolism of the chemicals,while the conventional pharmacokinetic models are'empirical' or 'data based' ones.In addition,the PBPK model is essential for the extrapolations.This review summaries the existing methodologies and techniques in PBPK model development,introduces chemical mixture's PBPK model,discusses PBPK model's applications in toxicology and health risk assessment,and in the final section,points out the current limitations and the possible future developments of the PBPK modeling approach.
