Purpose. Tight junctions have been suggested to be directly involved in the “barrier” and “fence” functions in cellular sheets, but little was known of their adhesion molecules until recently. Methods. This review discusses our current understanding of the molecular architecture of tight junctions, focusing on the recent discovery of two distinct types of tight junction-specific integral membrane proteins, occludin and claudins.
