Pharmacokinetics of single-dose intramuscular injection of Methotrexate for the treatment of ectopic pregnancy

OBJECTIVE To study the pharmacokinetics of single-dose intramuscular injection of Methotrexate in patients with EP(ectopic pregnancy)and calculate the pharmacokinetic parameters.METHODS After a single intramuscular dose(1 mg·kg-1)was given to nine patients with EP,the serum samples were withdrawn from 0 to 24 h and the blood drug concentration was assayed by fluorescence polarigation immunoassay.The pharmacokinetic parameters were calculated with 3p97 program.RESULTS The pharmacokinetics of single-dose intramuscular of methotrexate(MTX) in patients with EP conformed to two compartment with 1st order absorption.CONCLUSION The results would be helpful for the analysis of the factors contributed to the successful treatment rate.
