Objective:To explore the effect of Matrine combined with cisplatin on proliferation of A549 lung adenocarcinoma cells cultured in vitro,and explore its mechanism.Methods:A549 celline was treated with different concentration of Matrine and cisplatin,individually or in combination.And then cellular proliferation was evaluated by MTT method.VEGF expression in A549 cells in different concentrations of cisplatin and Matrine combined with cisplatin group was detected by RT-PCR analysis.Results:MTT assay showed that Matrine and cisplatin could also inhibit the proliferation of A549 cells in a dose and time-dependent manner in vitro(P0.05).Compared with Matrine and cisplatin individual drug groups,Matrine combined with cisplatin groups enhanced the growth inhibition in A549 cells.And the expression of VEGF mRNA was downregulated significantly in A549 cells treated by different concentrations of Matrine combined with cisplatin groups(P0.01) in a dose-dependent way.Conciusion:Matrine and cisplatin could inhibit the proliferation of A549 cell line in a dose and time-dependent manner,respectively.Matrine could increase the cytotoxic effect of cisplatin on A549 cell.It can be inferred that Matrine and cisplatin may inhibit lung cancer vascular growth attributable in part to downregulation expression of VEGF.
