Plasma Membrane-Cell Wall Adhesion is Required for the Responses of Mechanical Stress in Plant Cell

Objective: To study the morphological change of plant cell during normal culture after loading of mechanical stress and the role of plasma membrane-cell wall adhesion during the responses process. Methods: Plant cells were embedded in modified agarose MS medium and loaded for 24 hours, then cultured in culture room for several days. Observed by microscope everyday and make morphological analysis. Interruption of membrane-wall adhesion by mild plasmolysis and treated with RGD peptide. Treated cells were embedded, loaded and cultured the same way as above step. Results: Plant cell tend to expanded and multiplied perpendicular to the direction of main stress. Treated cells lost the ability of directional dividing witch perpendicular to the direction of main stress. Cells expanded and divided unordered. Conclusion: Plasma membrane-cell wall adhesion especially the RGD depended system is involved in the responses of mechanical stress in plant cell.
