摘要
Background
Group I metabotropic glutamate receptors (mGluRs), including mGluR1 and mGluR5, are G protein-coupled receptors. Signal transduction of mGluRs regulates chronic pain, emotion, cognition and other complex behaviors, and is a good candidate of drug targets in treatments. Scaffolding proteins Homer influences signal transduction of mGluRs by interacting with the mGluR5 intracellular C terminal TPPSPF motif through its EVH1 domain.
Objective
Since mGluR5 plays an essential role in pain, the role of mGluR5-Homer crosslinking on pain adjustment is reviewed.
Content
We summarized and separately expounded the structure of mGluR5-Homer complex and their regulation of pain.
Trend
It is more convincing that the expression of Homer1a alleviate pain via disassociating of mGluR5-Homer complex.
