Objective: To investigate the biological effect of cisplatin (CDDP) on melanoma Cloundman S91 cells. Methods: The techniques of MTT reduction assay, morphology examination, flow cytometry analysis were used. Apoptotic cells were labeled with TdT-mediated dUTP nick end labeling (TUNEL) method on cell smears. Results: cisplatin inhibited the growth of S91 cell line at the concentrations of 10-30 μmol/L. The cells treated with CDDP for 48 hours showed a typical apoptotic morphology and hypodiploid peak before G1 phase. Apoptotic index (AI) increased in a time-dependent pattern and concentration-dependent pattern. Conclusions: CDDP can induce melanoma Cloundman S91 cell apoptosis at the concentrations of 10-30 μmol/L.
