Objective To create the tissue enginering heart valve(TEHV) by seeding human umbilical vein endothelial cells( HUVECs )on decellular porcine aortic valve and investigate its physiological function.Methods The porcine aortic valve was decellularized using trypsin,EDTA,Triton X-100,RNase and DNase treatment. HUVECs were cultured and seeded on decelluar valve to create tissue engineering heart valve in vitro. Histology and growth condition of the cells were observed. Contents of t-PA,PAI-1,PGI_2,NO and ET-1 of endothelial cells were tested. Results The decellularization procedure resulted in complete removal of the cellular componentswhile the construction of matrix was maintained.The stress / strain curve between decellular valves and fresh valves were similar.The tissue engineering heart valve by seeding HUVECs on decellular porcine aortic valve in vitro was created successfully.Endothelial cells formed a continuous monolayer on the surface of valve and could secrete t-PA,PAI-1,PGI_2,NO and ET-1. Conculsion Tissue engineering heart valve can be created using HUVECs as seedingcells on the decellular porcine aortic valve,andendothelial cells has the normal secreting function.
