The effect of ultraviolet on cytokine secretion in human keratinocyte HaCaT cell line

Objective To observe the effect of UV on cytokine secretion in human keratinocyte HaCaT cell line, normal and stimulated by PMA and LPS respectively. Methods Expression of IL 1α,IL 2,IL 6 and IL 8 was detected by ELISA kit. Results Normal HaCaT cell secreted IL 1α,IL 2,IL 6 and IL 8 spontaneously. Stimulation by PMA and LPS induced increase in IL 6 and IL 8 level( P 0 01) and decrease in IL 1α level( P 0 01) in HaCaT cells. Acting on normal HaCaT cells, UV only increased the secretion of IL 8 distinctly( P 0 01). while changes were seen in IL 1,IL 2 and IL 6. Acting on HaCaT cells stimulated by PMA and LPS, UV increased the secretion of IL 1α,IL 6,IL 8. Conclusions The results indicated that the effect of UV on normal and stimulated HaCaT cells was different.
