Objective To study the mechanism of L929 cell death induced by Evodiamine in vitro.Methods Cytotoxicity of L929 induced by Evodiamine was determined by MTT method.Morphological changes of L929 cells under Evodiamine in- duction by inverted microscope and fluorescent staining.Apoptosis pathway was proved by LDH activity determination.Ef- fect of Evodiamine on cell cycle was studied by flow cytometry analysis technique.Results MTY method showed that Evodiamine of different concentrations could obviously inhibit L929 growth in dose-dependent manner.Evodiamine could induce L929 apeptosis,with the manifestation of apoptotic body and appearance of hypodiploid peak.But there was no typ- ical DNA trapezoid swimming band.Evodiamine of low dosage of 5μmol/L induced G_2/M stage arrest,while that of large dosage of 20 μmol/L induced G_0/G_1 stage.Conclusion Evodiamine of low dosage would induce G_2/M stage arrest while that of large dosage induce G_2/M_1 stage arrest in L929 cell apoptosis.
