Response surface analysis methodology(RSM) was used for optimizing ultrasound-assisted extraction of Tricholoma matsutake mycelium polysaccharides.Based on single-factor experiments, three independent variables, namely extraction time,water-solid ratio and ultrasonic power, were selected as casual factors. The experiments were arranged according to Box-Behnken central composite experiment design.Response surface analysis was applied to determine the effect of central composite experiment design.Response surface analysis was applied to determine the effect of casual factors on the yield of T.matsutake polysaccharides. The results showed that the optimum ultrasound-assisted extraction condition for T.matsutake polysaccharides was as follows:T.mastutake 0.1 g,extraction time 223 s,ultrasonic power 427 W and ratio of water to solid 69.7 ∶1(mL ∶g).The predicted best extraction yield was 14.52 %,while actual extraction yield was 14.33 %,with relative error 1.29 %,and the yield was 16.29 % by extracting twice.Anti-oxidative effect of T.matsutake polysaccharides was studied. The result showed that inhibition rates of polysaccharide on ·O-2 and ·OH could reach 19.32 % and 88.24 %,respectively.
