[Effect of electroacupuncture on Treg/Th17 balance in spleen and serum inflammatory factors in obese mice].

To observe the therapeutic effect of electroacupuncture(EA) on obese mice, and to explore the underlying mechanism of EA in treating obesity by focusing on the balance of regulatory T cells (Treg) and T helper 17 cells (Th17) and related inflammatory factors.C57BL/6J male mice were randomly divided into normal group, model group and EA group, with 10 mice in each group. The obesity model was established by feeding the mice with high-fat diet. Mice in the EA group was treated with EA at "Zhongwan"(CV12), "Guanyuan"(CV4), "Zusanli"(ST36) and "Fenglong"(ST40) for 20 min every time, 3 times every week, for a total of 8 weeks. The food intake and body weight of mice were observed and recorded, and Lee's index was calculated; the contents of interleukin 2(IL-2), IL-4, IL-6, IL-10, IL-17A, gamma interferon (IFN-γ) and tumor necrosis factor(TNF)-α in serum were detected by multiplex liquid chip quantitative technique; the levels of Treg and Th17 cells in mice spleen tissues were detected by flow cytometry; and the expression levels of foxhead box p3(Foxp3) and retinoic acid related orphan receptor γt(ROR-γt) mRNA in spleen were detected by real-time quantitative PCR.Compared with the normal group, the food intake, body weight, Lee's index, the contents of IL-2, IL-6, IL-17A, IFN-γ and TNF-α in the serum, and the percentage of Th17 and expression of ROR-γt mRNA in the spleen tissues were significantly increased (P<0.01, P<0.001), while the contents of IL-4 and IL-10 in the serum, the percentage of Treg and expression of Foxp3 mRNA in the spleen tissues were significantly decreased (P<0.001, P<0.01) in the model group. Compared with the model group, the food intake, body weight, Lee's index, the contents of IL-2, IL-6, IL-17A, IFN-γ, and TNF-α in the serum, the percentage of Th17 and expression of ROR-γt mRNA in the spleen tissues were significantly decreased (P<0.01), while the contents of IL-4 and IL-10 in serum, the percentage of Treg and expression of Foxp3 mRNA in the spleen tissues were significantly increased(P<0.01, P<0.05) in the EA group.EA may improve the obese state of mice by regulating the balance of Treg/Th17 in spleen and the expression of inflammatory factors in serum.目的:以调节性T细胞(Treg)与辅助性T细胞17(Th17)平衡及血清相关炎性因子为切入点研究电针治疗肥胖的潜在机制。方法:雄性C57BL/6J小鼠随机分为正常组、模型组、电针组,每组10只。采用高脂饮食建立小鼠肥胖模型,电针组选取“足三里”“丰隆”“中脘”“关元”给予电针治疗,20 min/次,每周3次,共干预8周。观察各组小鼠进食量、体质量并计算Lee’s指数;多重液相芯片定量技术检测各组小鼠血清中白细胞介素(IL)-2、IL-4、IL-6、IL-10、IL-17A、γ干扰素(IFN-γ)、肿瘤坏死因子α(TNF-α)的表达;流式细胞术检测各组小鼠脾脏组织中Treg、Th17细胞水平;荧光定量PCR法检测各组小鼠脾脏组织中叉头框蛋白P3(Foxp3)、维甲酸相关孤儿受体(ROR-γt) mRNA的表达水平。结果:与正常组相比,模型组小鼠的进食量、体质量、Lee’s指数明显升高(P<0.01,P<0.001);血清中IL-2、IL-6、IL-17A、IFN-γ、TNF-α含量升高(P<0.01),IL-4、IL-10含量降低(P<0.001,P<0.01);脾脏组织中Treg百分比及Foxp3 mRNA表达下降(P<0.01),Th17百分比及ROR-γt mRNA表达升高(P<0.01)。与模型组相比,电针组进食量、体质量、Lee’s指数明显降低(P<0.01);血清中IL-2、IL-6、IL-17A、IFN-γ、TNF-α含量明显降低(P<0.01),IL-4、IL-10的含量升高(P<0.01,P<0.05);脾脏组织中Treg百分比及 Foxp3 mRNA表达升高(P<0.05),Th17百分比及ROR-γt mRNA表达降低(P<0.01)。结论:电针可能通过调节肥胖小鼠脾脏Treg/Th17平衡及血清炎性因子的表达来改善其肥胖状态。.
