摘要
Objective To transfect HaCaT cells with adenovirus vector containing ERK-2 gene(Ad-ERK2) and to investigate the effects of ERK signal pathway on HaCaT proliferation induced by EGF.Methods The effects of EGF and PD98059 on HaCaT /transfected HaCaT proliferation were determined by 3H-TdR incorporation.Transfection ratio of adenovirus vector was detected by FACS and ERK-2 expression was identified by Western blot.Results 1 μg/L and 10 μg/L EGF promoted HaCaT proliferation,but the promoting effect of 100 μg/L EGF decreased.PD98059 inhibited the proliferation of HaCaT induced by EGF.Adenovirus vector efficiently transfected HaCaT,and the transfected HaCaT expressed ERK2 strongly.10 μg/mL EGF inhibited the proliferation of transfected HaCaT cells.Conclusion ERK signal pathway has both positive and negative effects on the proliferation of HaCaT cells induced by EGF.
