Objective To evaluate which digestive conditions could improve the yield of mouse pancreatic islets isolation.Methods Dif-ferent concentration of collagenase P was used by pancreatic intraductal injection,and the pancreas was digested for different times.After di-gestion,the islets were purified using discontinuous gradient centrifugation.The number,purity,and viability of the isolated islets were evalu-ated.Results The concentration of collagenase P and the time of digestion were important factors for efficacious isolation of islets.The con-dition of digestion by 1.0 mg/ml collagenase P for 25 min was optimal for islet isolation.Conclusion The collagenase concentration and the digestion time were important factors for islet isolation.Optimizing conditions for pancreas digestion can improve islet purity and viability of mouse islet isolation.
