Objective To observe the inhibitory effect of puerarin on aorta genetic expression of collage Ⅳ in experimental diabetic rats.Methods Diabetic rats were induced by intraperitoneal injection of streptozotocin.Rats were allocated to normal control group,model group,and three puerarin groups(20,40,80 mg·kg-1,i p)for 16 weeks.During and after the treatment,its general states(mind,physical quality),blood sugar,and glycosylated hemoglobin(HbA1c) alteration of tissue morphology via hematoxylin and eosin staining were observed after the aorta was taken out from the rats executed by narcosis;mRNA of collage Ⅳwas determined by in situ hybridization analysis.Results Diabetes mellitus and aorta lesion occurred in the four model groups.Puerarin could improve the general state,and decrease glycosylated hemoglobin(P0.05),the expression of collage Ⅳ mRNA in the aorta were significantly inhibited(P0.05),and the action of puerarin was proved to be related to the dose.Conclusions Puerarin shows preventing effect on aorta by inhibiting aorta genetic expression of collage Ⅳ.
