Objective To investigate the possible mechanism and the effect of xuebijing injection on tumor necrosis factor alpha(TNF-α) of the lung ischemia-reperfusion process.Methods 30 rabbits were randomly divided into five groups(n=6 each group): sham group(Group A),ischemia-reperfusion group(Group B),treatment group one day before surgery(Group C1),treatment group two days before surgery(Group C2),treatment group three days before surgery(Group C3).Left pulmonary artery was blocked by Berman balloon to creat pulmonary embolism model.2h later,50ml of physiological saline was bumped via the left femoral vein.Then the balloon was deflated to create the reperfusion model.before embolization,2h after embolization,reperfusion immediately,30min,2h,4h,respectively,antibody sandwich enzyme-linked immunosorbent assay(ELISA) was used to detect the blood concentration of TNF-αReverse transcription-polymerase chain reaction(RT-PCR) was used to detect the leukocyte TNF-α mRNA level 30 minutes after reperfusion in each group.Result Compared with the control,ischemia-reperfusion can improve TNF alpha levelsignificantly,Xuebijing injection can inhibit its expressiontimedependently.Conclusions Rabbit pulmonary embolism ischemia-reperfusion injury was related to change of the blood TNF alpha level.TNF alpha can aggravate ischemic reperfusion injury and Xuebijing injection can inhibit its expression to relieve the I/R injury.
