The content changes of aucubin,chlorogenic acid and PDG during heating in the barks and leaves of Eucommia ulmoides Oliv.

Objective:To study the content changes of aucubin,chlorogenic acid and pinoresinol diglucoside (PDG)in the barks and leaves of Eucommia ulmoides during heating by HPLC.Method:An HPLC method was es- tablished using Sunfire~(TM)C_(180 (5μm,4.6mm×250mm)column,the mobile phase consisted of acetonitrile-water (2:98),acetonitrile-0.4% phosphoric acid (13:87),acetonitrile-water (12:88),the flow rate was 1.0mL· min~(-1)and the detection wavelengths were 206,237,278nm.Results:The linear ranges of aucubin,chlorogenic acid and PDG were 2.168-34.688μg,1.2-19.2μg,0.964-15.424μg,respectively.The contents of the aucubin and chlorogenic acid in the leaves which were in boil water for 5 minites and then dried in 60℃,were much higher than those in the traditional processing products.However,the content of the aucubin in the barks which were dried in 80℃ was the highest in all,the contents of the chlorogenic acid and the PDG in the barks had no abosolutely change.Conclusion:The method can offer references for the processing and saving up Eucommia ulmoides.
