•The significance of cardiac Drd5 in the development of hypertrophic cardiomyopathy to heart failure via ROS over-generation by NADPH oxidase activation and mitochondrial dysfunction. •Drd5 myh6fl/fl-creERT2 mice as stable model maximally mimics the clinical phenotype of human HF, which fills a gap of mouse models in oxidative stress-induced HF.
