Objective To investigate the effect of advanced glycation end products(AGEs) on the proliferation of cultured rat vascular smooth muscle cells(VSMCs) and the inhibitory effect of aminoguanidine, pioglitazone, irbesartan on it.Methods Primary cultures of smooth muscle cells from rat aorta were exposed to AGEs of different concentrations and different times, and then exposed to AGEs(200mg/L) co-treatment with different doses of pioglitazone, aminoguanidine or irbesartan for 24 hours.3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assay was adopted for the quantification of the cells proliferation ratio. Results Cell proliferation in different AGE-BSA concentration medium was greater than that in nonglycated BSA medium(P0.05),except for that at 8 hours.Pioglitazone, aminoguanidine or irbesartan inhibited the AGE-BSA-induced cell proliferation of VSMCs(P0.05).Conclusion Aminoguanidine, pioglitazone or irbesartan may inhibit the AGE-BSA-induced cell proliferation of MSMC,and has good prospects in the treatment of diabetes and its vascular complication.
