OBJECTIVE: To review the relationship of transient receptor potential vanilloid receptor (TRPV) 5, 6 with bone metabolism. DATA SOURCES: An online search at PubMed database between January 1999 and July 2006 was undertaken to identify the related articles with the key word of TRPV, and the language was limited to English. STUDY SELECTION: After the primary check, literature including TRPV5 and TRPV6 was looked over for full-texts. Inclusive criteria: the study of TRPV5/6 and TRP super families. Exclusive criteria: the study of TRPV1-4. DATA EXTRACTION: Totally 106 related articles were collected, and 30 articles of them met criterions. DATA SYNTHESIS: TRPV5 and TRPV6 belong to the super-family of transient receptor potential channels. They are special epithelial-like Ca2+ channels. Recently, many studies proved that TRPV5 and TRPV6 are expressed in intestines and kidney, and play key roles in regulation of Ca2+ transport over cells. However, the reports about expressions of TRPV5 and TRPV6 in bone tissue are seldom, especially less about the bone metabolic mechanism. Therefore, this article reviews the relation TRPV5 and TRPV6 with bone metabolism. CONCLUSION: The role of TRPV5 and TRPV6 Ca2+ channels in bone metabolism provides the molecule therapy for the diseases associated with bone metabolism.
