Amphotericin B has been used to treat mycoses for decades and is one of the most prescribed drugs. A semi-solid formulation (emulgel) based on amphotericin B (AmB) and bacuri (Platonia insignis Mart.) butter, indicated in the treatment of cutaneous leishmaniasis, was analyzed, and a protocol for simple and rapid quantification of AmB was developed using ATR-FTIR spectroscopy. The method was based on the determination of AmB by measuring the area under the curve (AUC) of the band corresponding to the C – O stretching of the pyranose rings at 1040 cm−1 for the development of the calibration curve. The proposed method was validated according to the International Council for Harmonization, Validation of Analytical Procedures (ICH) guidelines regarding specificity, linearity, precision, and accuracy. The developed method was linear in the range of 1–6% (w/w) (R2 0.998) and met the validation parameters, confirmed by the low relative standard deviation values.
