Effects of IL-10 on TGF-β induced transdifferentiation of fibroblasts

Objective To explore the effects of IL-10 on TGF-β induced transdifferentiation of fibroblasts,multiplications and generation of collagen.Methods Human embryo lung fibroblasts were divided into 5 groups according to different treatment.Group A:treated with TGF-β at the concentrations of 5 ng/ml,Group B:treated with TGF-β at the concentrations of 5 ng/ml and IL-10 at the concentrations of 5 ng/ml,Group C:treated with TGF-β at the concentrations of 5 ng/ml and IL-10 at the concentrations of 15 ng/ml,Group C:treat with TGF-β at the concentrations of 5 ng/ml and IL-10 at the concentrations of 25 ng/ml,Group E:control group,culture dishes not containing fibroblasts.After 72 hour,MTT evaluated the situation of cell multiplication,hydroxyproline digestion examination detected the collagen generation and α-SMA expression examinated by immunocytochemistry.Results The α-SMA expression of fibroblasts in group B,group C,group D was significantly higher than that in group A(all P0.01),the α-SMA expression in group C was higher than that in group B(P0.05) and significantly lower than that in group D(P0.01).There were sifnificant differences in the collagen generation of fibroblasts between group B,group C,group D(all P0.01).The cell multiplication of group C was higher than that of group B(P0.05) and significantly lower than that of group D(P0.01).Conclution Different concentrations of IL-10 can reduce cell multiplication,collagen formation and α-SMA expression of fibroblast stimulated by TGF-β in vitro,its inhibitory effect depends on concentration.
