This work shows the metallation of DNA molecules containing Watson–Crick base pairs 7CG-C, where the guanine nucleobase has been replaced by its 7-deazapurine analogue. These deaza-DNA molecules can be initially assembled by using the Watson–Crick pairing rules, and the addition of silver(I) ions leads to the formation of silver–DNA molecules, comprising 7CG–AgI–C base pairs, that preserve their initial double helix B-conformation. This strategy, together with the generation of 7CA–AgI–T base pairs, can be employed to prepare silver–DNA systems by using their sequence programmability.
