摘要
ABSTRACT Five‐membered nitrogen‐containing heterocycles are crucial in medicinal chemistry because of their wide range of biological activities and presence in numerous pharmaceuticals and natural products. Important classes of these heterocycles include pyrroles, imidazoles, triazoles, tetrazoles, and pyrazoles, as well as their hybrid derivatives. These compounds can be synthesized using various synthetic approaches and exhibit diverse pharmacological properties, including anticancer, anti‐inflammatory, antitubercular, antimicrobial, and antioxidant activities. The biological efficacy of these heterocyclic scaffolds significantly influences their drug potency, pharmacokinetics, toxicity, and overall therapeutic spectra. Beyond pharmaceutical applications, these compounds are also useful in other fields such as agriculture. This review highlights the chemistry, synthesis, and biological applications of various five‐membered nitrogen‐containing heterocycles reported in the historical and recent literature, thereby providing a comprehensive overview of the importance of these compounds as key scaffolds in organic chemistry and medicinal chemistry. Furthermore, it emphasizes future perspectives and the ongoing evolution of research on nitrogen‐containing heterocycles, underscoring their potential for the development of novel therapeutic agents.
