Diagnosis and Therapy of Acute Myeloid Leukemia Abstract. Acute myeloid leukemia (AML) is a biologically complex and molecularly and clinically heterogeneous disease, and its incidence is increasing as the population ages. Cytogenetic anomalies and mutation testing remain important prognostic tools for tailoring treatment after induction therapy. Despite major advances in understanding the genetic landscape of AML and its impact on the pathophysiology and biology of the disease, as well as the rapid development of new drugs, standard treatment options have not experienced major changes during the past three decades. Allogeneic hematopoietic stem cell transplantation (HSCT) remains the best chance for cure. Here we review diagnosis and therapy of AML.Zusammenfassung. Die Akute Myeloische Leukämie ist eine biologisch komplexe und molekular und klinisch heterogene Erkrankung, deren Inzidenz mit dem Alter zunimmt. Zytogentische Anomalien und die Mutations-Testung sind wichtige prognostische Tools, um die Therapie nach der Induktions-Therapie zu steuern. Obwohl es deutliche Fortschritte im Verständnis der genetischen Veränderungen und deren Einfluss auf die Pathophysiologie und Biologie der Erkrankung gibt; und der Entwicklung neuer Medikamente; hat sich die Standard-Therapie in den letzten drei Jahrzehnten nur wenig verändert. Die allogene hämatopoietische Stammzell-Transplantation bietet die beste Chance auf Heilung. Dieser Artikel gibt eine Übersicht über die Diagnostik und Therapie der AML.
