Objective To study effect of benzo(a)pyrene[B(a)P] at different doses for different treating time on cell viability of human embryonic lung fibroblast(HELF) in vitro. Methods HELF cells at exponential growth phase in vitro planted in 96 wells microplates were treated by low doses of 0.018-10.000 μmol/ L and high doses of 15.6-125.0 μmol/ L B(a)P for 2, 4, 6, 24 h and 2, 4, 6, 24, 48, 72 h respectively. MTT assay was used to evaluate the effects of B(a)P on cell viability of HELF cells. Results Compared with the negative control, neither the effects on HELF cell viability of B(a)P at doses of 0.018-10.000 μmol/ L for 2, 4, 6, 24 h nor the effects at doses of 15.6-125.0 μmol/ L for 2, 4 h were observed. At the dose of 125.0 μmol/ L for 6 h, at the doses of 125.0?62.5?31.3 μmol/ L for 24, 48 h and at the doses of 15.6-125.0 μmol/ L for 72 h, cell viability of HELF were inhibited significantly. Conclusion Effects of B(a)P on cell viability of HELF were significantly dependent on the doses and the time.
