Objective To explore new preservation agent for lyophilization of red blood cells. The effects of trehalose and glucose on loading red blood cells were studied in the present study. Methods The red blood cells were loaded with 0,0.125,0.25,0.5 and 1 mol/L trehalose,glucose and combination of trehalose and glucose respectively in 37℃ for 6 hours. The levels of trehalose and glucose in the loaded red blood cells were analyzed respectively. Results There were not significant differences of trehalose and glucose levels in the loaded red blood cells among the three treatment groups (0.125,0.25,and 0.5 mol/L trehalose,glucose,combination of trehalose and glucose). However,the concentrations of trehalose and glucose in the red blood cells loaded with combination of trehalose and glucose (1 mol/L) significantly decreased than those of 1 mol/L trehalose or glucose treatment alone. Conclusion It suggested that loading with combination of trehalose and glucose (1 mol/L) had no significant influence on the concentrations of trehalose and glucose in the loaded red blood cells. The combination of trehalose and glucose was qualified for loading red blood cells for purpose of lyphilization.
