High-throughput experimentation (HTE) is generally understood to be the execution of many experiments in a parallel format using multi-well plates. Types of HTE experiments include parallel synthesis; HTE for reaction discovery; and HTE for reaction optimization. This chapter focuses on HTE for reaction discovery and optimization. It addresses common misconceptions and key advantages of HTE as a technique in synthetic chemistry. The chapter discusses key considerations and practical resources for typical HTE workflows, which are geared toward synthetic chemists with little or no experience with this technique. To illustrate how HTE workflows can be generalized and used for new discoveries, it then discusses two case studies in detail: The first study describes the development of a screening platform for Ni-catalyzed Suzuki–Miyaura couplings; the second study is a story of reaction discovery and optimization based on the authors' own experience.
