HPLC Method of Sialic Acid and Its Application on Purification of Sialic Acid

An HPLC method for determination of sialic acid was developed.Chromatographic conditions were as follows:separation column was ZORBAX SB C 18 ,mobile phase was an aqueous solution of sulphuric acid with pH3,the flowrate was1.0mL/min,the injection volume was5.0μL,and the detection wavelength was210nm.Good linear relation between the concentration of sialic acid and peak area was obtained,and the quality of correlation coefficientwas0.9999.This method was used to measure sialic acid duringthe puriˉfying process.Combined with mass spectrometry,it was confirmed that the dimer of sialic acid was a compoˉnent in hydrolysate of polysialic acid.
