OBJECTIVE:To establish a RP-HPLC method for the determination of Matrine,the active constituent in Radix Sophorae Flavescentis.METHODS:The determination of samples was performed on Waters C18(250mm× 4.6mm,5μ m) with methanol-acetonitrile-water-triethy lamine(35:20:55:0.02) used as the mobile phase.The detector was operated at 205nm,and matrine was added as the external standard.RESULTS:The linear range of Radix Sophorae Flavescentis was 0.102 4~ 4.096 0μg(r=0.999 9),and the average recovery of which was 98.92%,RSD=1.91%(n=6).CONCLUSION:The method was proved to be accurate,sensitive,reproducible and high in separation,and it can be use for the quality control of Radix Sophorae Flavescentis and compound preparation that contain Radix Sophorae Flavescentis.
