A comparison of the effect of degreasing from bone with alkali and acetone was made and the collagen production was measurated by the two methods.Finally the SDS-PAGE was used to analyze the purity of the collagen extracted.The results showed that the degreasing rate was the highest of 96.83% after dipping the bone powder in 1.0 mol/L NaOH solution for 3 days at 4 ℃,and the degreasing rate was the highest of 98.85% after dipping in the acetone for 3 days.The extracting collagen was the typical type Ⅰ collagen and its purity was 74.33%.Compared with alkali method,acetone method got better degreasing effect and collagen purity,and was more economic.
