Objective To develop an RPHPLC method with gradient dilution for simultaneous determination of chlorogenic acid and rutin in Lycium barbarum L.from different habitats.Methods The separation was performed on a Shim-pack VP-ODS 250 mm×4.6 mm(5 μm),acetonitrile/water/acetic acid(5∶94.5∶0.5)was used as the mobile phase A and acetonitrile/water/acetic acid(70∶29.5∶0.5)as the mobile phase B with gradient elution,the flow rate was 0.5ml·min-1,the detection wavelength was at 342 nm and the temperature was 30℃.Results The good linearities of chlorogenic acid and rutin were obtained over the ranges of 10~100 μg·ml-1(r=0.999 5 and r=0.999 8 respectively),the average recoveries were 98.6% and 99.0% respectively(n=5).The contents of chlorogenic acid and rutin in Lycium barbarum L.from different habitats were different.Conclusion The method is rapid and reproducible,the method can be used to determine chlorogenic acid and rutin in Lycium barbarum L.from different habitats.
