Objective By observing the changes of activity of protein kinase A(PKA) in granulation tissue and wound margin tissue of rabbit ear and speed of wound healing after using TGF-β1 to research the mechanism of TGF-β1 effecting. Methods TGF-pi was applied on the wound of rabbit ear.The PKA activity in the tissue from 1d, 3d, 6d, 11 -15d(i. e. time of wound epithelization) after wound formation and in normal skin was measured by phosphorus (32p) incoporation. The time of epithelialization was also observed. Results PKA activity in granulation tissue at wound epithelization and in wound margin tissue from 6d after wound formation was obviously elevated in comparing with that of normal skin(P0. 001 or 0.05) . TGF-β1 could not change PTK activity( P 0.05) and accelerated wound healing 1.5d(P0.05) . Conclusion The increase of PKA activity in wound healing might be related to healing rate. Unchangeable PKA activity by TGF-β1 suggested that PKA might not mediate the signal of TGF-β1 to accelerate wound healing.
