Objective: To study the anti-tumor mechanism of Wortmannin by observing the effect of Wortmannin on PANC-1 and CFPAC-1 cells' proliferation and apoptosis.Methods: The effects of Wortmannin on the proliferation and apoptosis of PANC-1 and CFPAC-1 cells were analyzed by methyl thiazolyl tetrazolium(MTT) assay,colony formation test and flow cytometry.Results: After treatment by Wortmannin for 24,48,72 h,the highest growth-inhibitory rate of CFPAC-1 cell was 10.6%?30.3%?18.9% respectively(P 0.05) and in a dose-dependent manner.In PANC-1 cell,cell growth was not influenced by Wortmannin.Wortmannin could inhibit the colony formation of both cell lines,but more significant in CFPAC-1 cell.In both cell lines,apoptosis was not observed.Conclusion: Wortmannin has the anti-tumor effects by suppressing cells proliferation through inhibiting the PI3K/Akt signal pathway.
