AIM:To study the pharmacokinetics of fluvastatin and glimepiride and their pharmacokinetic interaction in human.METHODS:According to Latin square design,12 male volunteers were randomized into six groups,and took fluvastatin(40 mg·d~(-1),for 8d),glimepiride(4 mg·d~(-1),for 7 d)or fluvastatin(40 mg·d~(-1),for 8 d)+glimepiride(4 mg d~(-1),for 8d).Plasma concentration of fluvastatin and glimepiride were measured by HPLC,respectively.DAS 2.0 programe was used to get pharmacokinetic parameters.RESULTS: When two drugs taken simultaneously,the pharmacokinetic parameters of fluvastatin showed no significant changes,but c_(max)~(ss),c_(min)~(ss),c_(av)~(ss),AUC_(0~24)~(ss),AUC_(0~∞)~(ss),t_(1/2)and MRT of glimepiride increased(P0.05)while CL (F)and DF decreased(P0.05).CONCLUSION:Since fluvastatin inhibits the metabolism of glimepiride in liver,plasma concentration of glimepiride should be monitored to prevent the occurrence of adverse drug reactions.
