Hirudin has been intensively investigated for research and therapeutic purpose. It's one of the most powerful anticoagulants. A specific extraction method from the salivary gland secretions of the alive leechs, Hirudinaria manillensis, had been applied. The purified hirudin can be obtained by precipitation procedures followed by an anion exchange chromatography on DEAE-C52 and a gel filtration chromatography on Sephadex-G25. The hirudin was analyzed by the method of titration with thrombin. The hirudin activity in salivary gland secretions is 5 AT-U/mL.
