This study describes a new method for determining tramadol hydrochloride in pharmaceutical ampoules that used Molecularly Imprinted Solid-Phase Extraction (MISPE) as a sample extraction technique. Molecularly Imprinted Polymer (MIP) was prepared by using 1-vinylamidazole as the functional monomer, trimethylolpropane trimethacrylate as the cross-linker and tramadol hydrochloride as the template molecule. The new imprinted polymer was employed to extract tramadol hydrochloride from pharmaceutical samples using solid-phase extraction (SPE) as a sorbent. The spectrophotometric method was measured using a UV-Vis spectrophotometer in the 200–400 nm range. In methanol media, the standard and sample solutions were prepared and The maximum absorption of tramadol hydrochloride was found at 272 nm in the UV absorption spectrums that were observed. Several parameters affecting the polymer’s extraction efficiency have been evaluated. The optimum conditions for MIP cartridges were investigated. The limits of detection (LOD) and quantification (LOQ) were 0.62 and 1.87 µg /mL, respectively. Relative Standard Deviation (RSD%) was found to be ˂ 2%. This method was successfully utilized for ampoule forms, and the results were thoroughly validated and proven to be durable.
