An analytical method for the determination of DNA was established by spectrophotometry, based on the decrease of absorbance at maximum absorption wavelength for crystal violet(589 nm) with addition of calf thymus DNA. The degree of decrease of the absorbance is linear with the amount of DNA. The determination conditions were optimized. The detection limit of 19.5 μg/L and linear range of 0~5 mg/L for the method were found. The method is simple, rapid and selective, and was applied to analysis synthetic sample with satisfactory.
