OBJECTIVE To elucidate the mechanism underlying the protective effects of Chinese herb baicalin and gardenoside on oxygen-glucose deprivation and reperfusion injury. METHODS SH-SY5Y cell were subjected to the oxygen-glucose deprivation and reperfusion injury, and the effects of baicalin and gardenoside on neurocytes were examined. RESULTS 8 hours of oxygen-glucose deprivation plus 12 hours of reperfusion led to severe injury to neurocytes. The activity of mitochondria decreased significantly, and the percentage of apoptotic cells increased significantly. It was also observed that baicalin and gardenoside could significantly alleviate this injury. CONCLUSION Ba-icalin and gardenoside had direct protective effects on neurons subjected to oxygen-glucose deprivation and reperfusion injury, which might be one of the mechanisms underlying its therapeutic effects on oxygen-glucose deprivation and reperfusion injury.
