Objective:To investigate the alteration of cytokine expression after immunosuppressive therapy,analyze their clinical significance in patients with aplastic anemia(AA),and explore the possibility of therapeutic efficacy on cytokine IL-8.Method:Amount of IL-8 in peripheral blood and bone marrow was measured by real time PCR methods. Changes of CD4/CD8 positive cells were analyzed by flow cytometery.Result:The level of cytokine IL-8 in peripheral blood and BMP of aplastic anemia patients was significantly increased than that in the normal control,and the ratio of CD4/CD8 reversed. The level of IL-8 in acute AA (typeⅠtypeⅡ) was higher than that in chronic AA. The ALG+CsA+male hormone therapy had been proved to be more effective than CsA+male hormone therapy. The total efficacy rate was 82.3% and 69.3%,and recovered CD4/CD8 ratio. The ALG+CsA+male hormone had a better effect in acute AA (typeⅠtypeⅡ). There was a significant correlation between the alteration of IL-8 and CD4/CD8 with therapy efficacy. Real time PCR was positively correlated with ELISA,and more easily detected and quantized.Conclusion:ALG allied CsA is effective in acute and chronic aplastic anemia therapy. Cytokine changes of IL-8 may be used as a serveillance marker in diagnosis and treatment in aplastic anemia.
