Objective: To investigate plasmid pSecTag2HygroC/CD13-1/RFP transfected into 293 cells by lipofectamine 2000.Methods: The plasmid pSecTag2HygroC/CD13-1/RFP was transfected into 293 cells by lipofectamine 2000,then determined by RFP fluorescence.The culture fuild of transfected 293 cells was collected and concentrated.The embranin and plasmosin of transfected 293 cells were extracted to verify whether the transfection cells could express the interest protein using Western blot.Results: ① The expression of RFP in 293 cell was observed under fluorescence microscopy.② The result of Western blot showed that the plasmosin of transfected 293 cells(about 62kDa) was expressed stably.Conclusion: Through cell transfection with lipofectamine 2000,plasmid pSecTag2HygroC/CD13-1/RFP can be transfected successfully into 293 cells,which would be a firm foundation for further studies on this monoclonal antibodie applied in clinical therapy.
