Determination of gastrodin contents in wild and cultivated Gastrodia elata from different regions of Chongqing by RP-HPLC.

[Objective] To compare the gastrodin contents in wild and cultivated Gastrodia elata from different regions of Chongqing.[Method] After ultrasonic extraction,gastrodin content in G.elata was detected by RP-HPLC.The mobile phase was acetonitrile-0.05% phosphate solution (V/V,3:97);chromatographic column was DiamonsilC18 column (250 mm × 4.6 mm,5 μm);detection wavelength was 220 nm;flow rate was 1 ml/min;and column temperature was 30 ℃.[Result] Gastrodin content varied greatly in different production areas,and there were relatively great differences between cultivated and wild G.elata.Gastrodin content in wild G.elata was relatively higher than that in cultivated G.elata.Among them,cultivated G.elata in Chengkou and wild G.elata in Yunyang had relatively high content of gastrodin;while cultivated G.elata in Wushan showed relatively low content of gastrodin.[Conclusion] The gastrodin contents in cultivated and wild G.elata in Chengkou were far higher than the national standard.Therefore,it was completely feasible to establish planting base for G.elata in Chengkou that complies with the requirements of the national GAP.
